Principle of the Assay: This assay employs the direct competitive inhibition enzyme immunoassay technique. Antibody specific for HBD has been pre-coated onto a microplate. Standards and samples are pipetted into the wells with biotin-conjugated HBD. A competitive inhibition reaction is launched between HBD (Standards or samples) and biotin-conjugated HBD with the pre-coated HBD antibody. After washing, avidin conjugated Horseradish Peroxidase (HRP) is added to the wells. Following a wash to remove any unbound reagent, a substrate solution is added to the wells and color develops in opposite to the amount of HBD bound in the initial step.
